Abstract A symbiotic dialogue between the microbiota and host is essential to promote protective immune responses in skin. Integrated retroviral elements, or retrotransposons (RTNs), comprise up 45% of human genome. We previously found that skin are dependent on reactivation keratinocyte-intrinsic sensing RTNs. Here, we explored possibility RTNs may also represent a source antigens recognized by adaptive system. first established model sterile injury, where topical application mild detergent reactivated RTNs, recruited CD8 T cells an RTN-dependent manner. RNAseq analysis RTN expression revealed Langerhans (LCs) have most abundant amongst dendritic cells.
